Here's why you have to use black as the bottom layer. A layer's blending mode (Hard Light) determines how its pixels (the colors you will be putting on the layer) blend with the pixels underneath (black). If you look at the two pictures below I made a layer and filled four sections with a different color. Then I added a new layer above it and set the blending mode for the layer to Hard Light. When I made a line with the brush tool on the Hard Light layer you can see how the two colors I used, pink and blue, blended differently with each of the colors underneath. There will be a point in the signature where it won't matter anymore if your marbles are above the black layer in the layers palette. At that point if you choose you can add another layer with a different color. I personally think they look best with black.

Select the brush tool and make sure your settings are the same as in the picture. Choose this brush from the brushes palette. If you don't have this brush choose a round brush and change the settings to the ones in the picture.

On the blobs layer you are going to click down to make your blobs. You don't need to put your colors where I put mine nor do you have to use the same colors. I'm giving you the colors' settings so if you do like how certain colors look in the final marbles you'll be able to reproduce them. To get any of these colors click on the foreground color's box in the toolbox. In the Color Picker window enter the settings with the red outline and click OK. To save any of the colors in your swatches palette check out this thread, Swatches palette. These are the colors I used...
